Epidemiology and Pathogenesis of Dementia

Abstract
The prevalence of dementia is expanding rapidly, particularly in countries with low and middle incomes. Age-standardized prevalence for those aged ¡Ã60 years, 5?7% in most world regions, and it was estimated that 35.6 million people lived with dementia worldwide in 2010, with numbers expected to almost double every 20 years, to 65.7 million in 2030 and 115.4 million in 2050. In South Korea, the prevalence for those aged of dementia ¡Ã60 years in 2012. It is expected to 12.7 million in 2030 and 27.1 million in 2050. The most influential theory to explain the pathogenesis of Alzheimer¡¯s disease (AD) has been the ¡°Amyloid Cascade Hypothesis¡±. The hypothesis proposes that the deposition of ¥â-amyloid which is producted from amyloid precursor protein by ¥â secretase, and ¥ã secretase is the initial pathological event in AD leading to the formation of senile plaques and then to neurofibrillary tangles death of neurons, and ultimately dementia.
